style="FONT-SIZE: 14pt">Un enfoque racional para el descubrimiento de drogas se
basa en estudios estructurales para identificar blancos moleculares, en
particular componentes xenobióticos para el hospedero humano. En <I
style="mso-bidi-font-style: normal">Trypanosoma cruzi</I> hemos identificado
algunas de esas estructuras singulares. El lipopéptidofosfoglicano (LPPG)
definido luego como glicoinositolfosfolípido (GIPL) es la molécula de superficie
mas abundante en epimastigotes y tripomastigotes metacíclicos.<SUP>1 </SUP>En
esta biomolécula se pueden señalar <o:p></o:p></SPAN></P>
<P class=MsoNormal style="MARGIN: 0cm 0cm 0pt; TEXT-ALIGN: justify"><SPAN
style="FONT-SIZE: 14pt">tres estructuras ausentes en mamíferos : la
galactofuranosa (Gal<I style="mso-bidi-font-style: normal"><SUB>f</SUB></I><SUB>
</SUB>), el acido amino etilfosfónico (AEP) y la inositolfosfoceramida (IPC). La
biosíntesis de Gal<I style="mso-bidi-font-style: normal"><SUB>f</SUB></I> ha
atraído el interés de muchos grupos principalmente porque es también<SPAN
style="mso-spacerun: yes"> </SPAN>un componente crítico de <I
style="mso-bidi-font-style: normal">Mycobacterium tuberculosis. </I>Las dos
enzimas involucradas en su incorporación:<SPAN style="mso-spacerun: yes">
</SPAN>la mutasa que convierte UDP-</SPAN><SPAN
style="FONT-SIZE: 14pt; FONT-FAMILY: Symbol">a</SPAN><SPAN
style="FONT-SIZE: 14pt">-D-Galp en UDP-</SPAN><SPAN
style="FONT-SIZE: 14pt; FONT-FAMILY: Symbol">a</SPAN><SPAN
style="FONT-SIZE: 14pt">-D-Galf y una galactofuranosil transferasa han sido
clonadas. Hemos descripto la primera síntesis química de
UDP-<o:p></o:p></SPAN></P>

style="FONT-SIZE: 14pt">[6-<SUP>3</SUP>H]-</SPAN><SPAN

</SUB></I>, sustrato para estudios de inhibidores de <st1:PersonName w:st="on"
ProductID="la Galf">la Gal<I
style="mso-bidi-font-style: normal"><SUB>f</SUB></I></st1:PersonName>
transferasa independientemente de la mutasa.<o:p></o:p></SPAN></P>

style="mso-spacerun: yes"> </SPAN>junto con <st1:PersonName w:st="on"
ProductID="la Galp">la Gal<I
style="mso-bidi-font-style: normal">p</I></st1:PersonName> está presente en
mucinas del linaje 1 mientras que cepas del linaje 2 , involucradas en la
infección humana contienen sólo Gal<I style="mso-bidi-font-style: normal">p.</I>
Dado que las mucinas son las aceptoras de ácido siálico en la reaccion de
trans-sialidación, importante en la infección, hemos sintetizado los
oligosacáridos naturales para poder correlacionar su estructura con la habilidad
para actuar como sustratos.<SUP>2</SUP> <o:p></o:p></SPAN></P>

drogas. <st1:PersonName w:st="on" ProductID="La IPC">La IPC</st1:PersonName> es
un constituyente importante de <I style="mso-bidi-font-style: normal">T.
cruzi<SPAN style="mso-spacerun: yes"> </SPAN></I>en los tres
estadíos,<SUP>3<SPAN style="mso-spacerun: yes"> </SPAN></SUP>a diferencia
