peroxisome proliferators-activated receptors (PPARs) are dietary lipid sensors
that regulate fatty acid and carbohydrate metabolism.<SPAN
style="mso-spacerun: yes"> </SPAN>There are three known PPAR receptors:
alpha, gamma and delta.<SPAN style="mso-spacerun: yes"> </SPAN>Activation
of PPAR alpha results in hypolipidemic effects, activation of PPAR gamma results
in antidiabetic effects.<SPAN style="mso-spacerun: yes"> </SPAN>By
contrast, the therapeutic potential of PPAR delta receptor is unknown, due in
part to the lack of selective ligands. This presentation is going to show the
use of combinatorial chemistry and structure-based design that resulted in the
development of a potent and selective PPAR delta agonist: GW501516.<SPAN
style="mso-spacerun: yes"> </SPAN>This compound increases HDL (good
cholesterol) and reduces the levels of LDL (bad cholesterol), fasting
triglycerides, and fasting insulin.<SPAN style="mso-spacerun: yes">
</SPAN>GW501516 is now in Phase II clinical trials in USA for the treatment of
lipid metabolism disorder<o:p></o:p></SPAN></P></SPAN></DIV>
