<P><SPAN lang=en-us><FONT face=Arial>Research in</FONT></SPAN><SPAN
lang=en-us><B> <FONT face=Arial color=#367889>Dr. David Colman's</FONT><FONT
face=Arial></FONT></B> <FONT face=Arial>laboratory deals primarily with the
interactions between cell adhesion molecules. One model we use is the vertebrate
myelin sheath, which is a broad, flattened, self-adherent membrane that
surrounds axons and allows for the rapid conduction of the nerve impulse. The
sheath is an excellent model for studying fundamental questions about plasma
membrane biogenesis in the nervous system. How the almost crystalline
ultrastructure of the myelin sheath is assembled is a fascinating problem, and
the examination of the molecular interactions by which the sheath is formed is a
major focus of the laboratory. Studies on the biochemistry, molecular structure
and most recently the evolutionary origins of the myelin proteins are providing
intriguing insights into their general properties and their actual roles in the
formation and maintenance of the sheath.</FONT></SPAN></P>
<P><SPAN lang=en-us><FONT face=Arial>In other studies, we are focusing on the
adhesion mechanisms by which intercellular junctions, and in particular the pre-
and post-synaptic membranes in the CNS, are held together. We have found that
cadherins, calcium dependent adhesion molecules that operate in all epithelial
cells, are important components of the CNS synaptic junctions. In fact, it is
possible that the differential adhesive specificities of individual cadherins
may be responsible in large measure for the high degree of specificity of
synaptic connections in the CNS. </FONT></SPAN></P><BR>
