"Genomic repositioning of the major ribosomal protein locus alters growth
rate and host-invasion capacity of Vibrio cholerae."

It is poorly understood how gene order within the bacterial chromosome
influences cell physiology. In silico approaches revealed that ribosomal
protein genes locate near the replication origin in fast-growing bacteria.
Here, using novel recombination tools, we created a set of Vibrio cholerae
strains in which we relocated the S10-spec-α locus (S10), bearing
half of ribosomal protein genes, to different genomic positions. These
isogenic strains showed different phenotypes, since growth rate and
infectivity varied as a function of the distance to the origin of
replication. This was correlated with a reduction of S10 dosage and
expression. Returning S10 to its original location restored the original
growth rate as well as the host-invasion capacity, demonstrating that S10
positioning is essential for both phenotypes. We conclude that S10
positioning links genome structure with cell physiology and may be
important for the ecological success of V. cholerae.
